Expression of recombinant proteins from chimeric genes, engineered to containepitopes of the extracellular ligand binding region of the human lutropinreceptor and carboxy terminal region of hcG-β-CTP

Expression of His-tagged recombinant protein from a chimeric gene consisting of the extracellular ligand-binding domain (ECD) of hLH-R and the carboxyl terminal peptide (CTP) of hCG-β is described. Structural integrity of the chimera protein was confirmed by liquid chromatography-tandem mass spectrometry, by Western Blotting using antibodies to LH-R and hCG-β as well as against anti-6xHistidine and in an immunoassay. The immunized mice generated specific functional antibodies which bound both LH-R and hCG. Serum progesterone was suppressed in immunized mice while the estradiol levels did not change from that of the control mice.
